Anti-inflammatory activity of 5 cassaine diterpenoids isolated from the leaves of Erythrophleum fordii

Abstract

In this study, 5 cassain diterpenoids as erythrofordin A (1), erythrofordin B (2), erythrofordin C (3), pseudo-erythrosuamin (4) and erythrofordin U (5) are isolated from the leaves of Erythrophleum fordii Oliver collected in Quang Nam province. The chemical structures of these compounds are identified by 1H and 13C NMR and mass spectrum MS in comparasion with reported data. The anti-inflammatory activity of these compounds is evaluated against lipopolysaccharides (LPS)-induced nitric oxide production in RAW264.7 cells in vitro. Among them, erythrofordin B (2) shows significant inhibitory activitiy against the LPS-induced nitric oxide production with an IC50 value of 12.5  1.8 μM. Erythrofordin B suppresses both cyclooxygenase-2 and inducible nitric oxide synthase expressions in protein levels. These results indicate that erythrofordin B (2) may exert anti-inflammatory activity though NO production inhibition.
